- ·Bring sheet music in the correct key. An accompanist will be provided. No a cappella auditions.

The role of the Voice of Audrey II has been pre-cast.
- ·SEYMOUR KRELBORN (male, 20s–30s) — A self-doubting, overworked florist's clerk and the improbable hero. Sincere and well-intentioned but taken for granted because of his clumsy ways and poor social skills. In love with Audrey. Any ethnicity.
- ·AUDREY (female, 20s–30s) — All heart beneath her perfect hair and suggestive outfits. Sweet and vulnerable, with beautiful dreams for the future despite her abusive situation. Any ethnicity.
- ·MR. MUSHNIK (male, 50s–60s) — The weathered owner of the failing flower shop and Seymour's boss. Hungry for profit, gluttonous, and manipulative. Any ethnicity.
- ·ORIN SCRIVELLO (male, 30s–40s) — An egotistical dentist with sadistic tendencies and Audrey's abusive boyfriend. Also plays other larger-than-life characters; requires versatility and strong comic timing. Any ethnicity.
- ·URCHINS: CRYSTAL, RONNETTE, AND CHIFFON (female, Black, late teens–early 20s) — The street urchins and occasional Greek Chorus. Young, energetic, street smart, and the only ones with a grip on reality. Strong singers and movers.
- ·THE PLANT (AUDREY II) — Puppet. An anthropomorphic cross between a Venus flytrap and an avocado with an appetite for human flesh. Puppeteer doubles as other sizes of the plant.
